RPMI 1640 (Roswell Park Memorial Institute) was first used to culture human leukemia cells in suspended and monolayer media. This medium is suitable for culturing different types of mammalian cells including Hela, Jurkat, MCF-7 PC12, PBMC, astrocytes and carcinoma. The unique feature of RPMI 1640 culture medium is having glutathione as a reducing agent as well as large amounts of vitamins. This culture medium contains biotin, vitamin B12, and para-aminobenzoic acid, which are not present in EEM (Eagle’s Minimal Essential Medium) or Dulbecco’s Modified Essential Medium (DMEM). In addition, inositol and choline are present in large quantities in this culture medium. The RPMI 1640 medium does not contain any proteins, lipids or growth factors. Therefore, this culture medium requires supplementation, which is usually 10% bovine fetal serum (FBS). In this culture medium, sodium bicarbonate buffer (with a concentration of 2 g / l) is used and therefore to maintain its physiological pH requires 5-10% of crane dioxide in the environment. This product (BI-1006) specifically contains glutamax / L-glutamine, 15 mM HEPES, sodium bicarbonate and phenol red. To better maintain the physiological pH at different concentrations of carbon dioxide, HEPES (hydroxyl-piperazine ethane sulfonic acid) solution was added to the medium as a zoetronic chemical buffering agent.
